摘要:
目的  探讨环境雌激素多氯联苯同系物PCB153对发育过程中小鼠胚胎细胞凋亡的影响。  方法  于体外施予孕8.5 d小鼠胚胎不同浓度PCB153,应用植入后全胚胎培养技术培养胚胎;采用原位TUNEL技术观察胚胎细胞凋亡。  结果  PCB153对胚胎发育具有明显影响且呈剂量-效应关系,且随PCB153浓度增加,凋亡细胞比值上升。  结论  PCB153能诱导胚胎细胞凋亡,这与其致胚胎发育异常有关,可能是PCB153的致畸作用机制之一。
Abstract:
Objective    To explore the effects of polychlorinated biphenyl congener (PCB153) an environmental estrogen,  on embryonic development and its mechanism.     Methods    Mouse embryos of gestational day 8.5 were exposed to different doses of PCB153 ex vitro. Post-implantation whole embryo culture (WEC) technique was used to investigate the effects of PCB153 on growth and development. The terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end labelling (TUNEL) technique was used to detect apoptosis in embryonic tissue sections.     Results    PCB153 had obvious effects on the development and growth of embryos in a dose-dependent manner; The ratio of apoptotic cells was increased with increased doses of PCB153.     Conclusion    PCB153-induced apoptosis is involved in mouse embryo anomalies.
