摘要:
目的     比较活性氧、线粒体膜电位在线粒体DNA缺失肝癌细胞株SK-Hep1(ρ0SK-Hep1)与亲本细胞中的差别。     方法     采用溴化乙锭诱导制备ρ0SK-Hep1细胞株;荧光探针标记、流式细胞计数及激光共聚焦显微镜方法检测细胞内活性氧和线粒体膜电位。     结果     ρ0SK-Hep1细胞内活性氧荧光强度显著高于其亲本SK-Hep1细胞株(P<0.01),ρ0SK-Hep1肝癌细胞平均计数35.5,而SK-Hep1肝癌细胞平均计数15.9。ρ0SK-Hep1细胞线粒体膜电位显著低于亲本细胞(P<0.01),ρ0SK-Hep1细胞平均计数55.0,而SK-Hep1细胞平均计数65.9。     结论     线粒体DNA缺失后细胞内活性氧水平增加,而线粒体膜电位有所减低。
Abstract:
Objective     To observe the differences of the reactive oxygen species (ROS) and mitochondrial membrane potential (ΔΨm) in mitochondrial DNA-depleted SK-Hep1 and its parent cells.       Methods     ρ0SK-Hep1 cell line was generated by treating SK-Hep1 with ethidium bromide. Flow cytometry (FCM) and laser scanning confocal microscopy were used to detect ROS and ΔΨm.       Results     Fluorescence intensity of ROS in ρ0SK-Hep1 cells was significantly higher than that of the parent cells (35.5 vs 15.6), and the ΔΨm was lower in ρ0SK-Hep1 cells (55.0 vs 65.9).        Conclusion     ρ0SK-Hep1 cells showed elevated ROS and lower ΔΨm. These changes might contribute to the variation of phenotypes.
