目的    研究三羟异黄酮(Genistein,Gen)对人乳腺癌细胞系体外增殖的影响及其与端粒酶的关系。    方法    体外培养雌激素受体阳性(ER+)和阴性 (ER-)乳腺癌细胞MCF-7和MDA-MB-231, MTT法检测细胞增殖的影响;RT-PCR法观察端粒酶逆转录酶TERT基因mRNA的表达;免疫细胞化学染色观察NF-κB p65的核移位。    结果    雌二醇(Estradiol,E2)单独作用时,促进MCF-7细胞增殖、TERT mRNA表达和NF-κB p65的核移位,但对MDA-MB-231细胞无明显影响。低剂量的Gen对MCF-7细胞的单独作用与E2类似,与E2联合作用时,MCF-7细胞无明显变化;高剂量的Gen单独或联合E2作用于 MCF-7细胞以及MDA-MB-231细胞,均表现为抑制细胞增殖、TERT mRNA表达和NF-κB p65的核移位。    结论    Gen对两种乳腺癌细胞MCF-7和MDA-MB-231增殖的不同影响与雌激素受体表达状态及E2存在与否有关,并可能是通过调节TERT的mRNA表达、NF-κB p65的核移位来调控细胞端粒酶活性的。
Abstract:
Objective    To study the effects of Genistein (Gen) on the proliferation of human breast cancer cell lines in vitro and the relationship with telomerase.     Methods    Human breast cancer cell lines MCF-7 (ER+) and MDA-MB-231(ER-) were cultured in vitro. MTT assay was used to determine the effects of Gen and Estradiol (E2) on the proliferation of the two cell lines. RT-PCR was conducted to detect the expression of TERT mRNA and  immunocytochemistry assay was used to observe the nuclear translocation of NF-κB.     Results    E2 promoted the proliferation of MCF-7 cells (P<0.01), the expression of TERT mRNA (P<0.01) and the nuclear translocation of NF-κB, but had no effect on MDA-MB-231 cells. The effect of Gen alone on  MCF-7 in low dose was similar to E2, and the effect of Gen in combination with E2 was similar to the control. For both cell lines, in high dose, Gen inhibited the proliferation(P<0.01), the expression of TERT mRNA (P<0.01) and the nuclear translocation of  NF-κB p65 when used alone or in combination with E2.     Conclusion    The varied effects of Gen on MCF-7 and MDA-MB-231 cells are related to the status of ER expression and the presence or absence of estrogen, and the underlying mechanisms of these effects may be through regulating the TERT expression and nuclear translocation of NF-κB p65.
