目的 探讨小鼠烧伤早期干扰素诱导蛋白P56、IFN-β、IFN-α mRNA在肺、肝、脾、肾组织中的表达,以及烧伤后给予麻醉剂或KPV对其表达的影响。 方法 选取健康昆明鼠40只,用随机排列表法分为4组:正常对照组、单纯烧伤组、烧伤+KPV组(简称KPV组)、烧伤+麻醉组(简称麻醉组),烧伤后3 h取肝、肺、肾、脾组织,提取RNA,用两步法RT-PCR检测P56、IFN-β、IFN-α mRNA的表达。 结果 P56 mRNA在肝、脾、肺、肾组织中都有表达,且表达丰度较〖JP2〗高。在肾组织中烧伤组和KPV组表达显著高于正常组(P<0.05),麻醉组P56 mRNA表达与正常组相似;IFN-β mRNA在肝组织中不表达,而在脾、肺、肾组织中均有表达,但表达量不高,在肺组织中烧伤组和麻醉组表达显著增加(P<0.05),KPV组表达略微降低;IFN-α mRNA在所有已测脏器中都有表达,在肾组织中烧伤组和KPV组较正常组表达显著增加(P<0.05),麻醉组增加不明显。 结论 烧伤后干扰素表达增加,诱导P56表达增加,说明P56可能与早期创伤应激不良有关。
Abstract:
Objective To investigate the expressions of P56, IFN-β, IFN-α mRNAs and the effects of KPV, a COOHterminal tripeptide sequence of alphamelanocyte stimulating hormone [alpha-MSH(11-13)], and anaesthesia on the expressions in severe burn injured mice.  Methods Eighty mice were randomly and equally divided into 4 groups: control, burn (TBS15%Ⅱ), burn+KPV (injection of 40 μg per animal immediately after burn), and burn+anaesthesia (injection of 0.4 mg/kg diazepam and 10 mg/kg ketamine immediately after burn). All animals were sacrificed in 3 h after burn respectively. The expressions of P56, IFN-β, and IFN-α mRNAs in liver, spleen, lung and kidney tissues were detected with RT-PCR.  Results P56 was expressed highly in liver, spleen, lung, and kidney in normal control group, and was obviously increased in kidney in burn group and burn+KPV group, but that in burn+anaesthesia group had no significant difference with that in normal control. IFN-β was not expressed in live and lightly expressed in spleen, lung and kidney, and its expression in lung was significantly increased in burn group and burn+anaesthesia group, but mildly decreased in burn+KPV group. IFN-α mRNA was expressed in all detected organs in normal control, and highly expressed in kidney of burn group and burn+KPV group, but not in burn+anaesthesia group.  Conclusion These results indicate that the increased IFN expressing induce the up-regulation of P56, suggesting which may be involved in the disorder of early post trauma stress.
