摘要:
目的 探讨当归多糖(Angelica polysaccharides, APS)动员的造血干/祖细胞(hematopoietic stem/progenitor cell, HSPC)对造血衰竭小鼠造血功能重建的作用。 方法 将APS动员的雄性BALB/c小鼠的外周血单个核细胞(peripheral blood mononuclear cells, PBMC)静脉输注给受到8.5 Gy 60Co γ射线照射的雌性同系受体小鼠;观察受体小鼠外周血白细胞(WBC)、血红蛋白(HGB)、血小板(PLT)的变化及30 d存活情况;采用PCR方法鉴定其造血重建的来源。 结果 APS组受体小鼠WBC、PLT、HGB、30 d存活数均明显高于对照组和NS组(P<0.05);对APS组存活小鼠进行Y染色体PCR分析,证实雌性受体小鼠重建的造血细胞来自雄性供体。 结论 APS动员的小鼠外周血HSPC移植后能够重建造血衰竭小鼠长期造血。
Abstract:
Objective  To study hematopoiesis reconstruction in mice of hematopoietic aplasia by transplanting hematopoietic stem/progenitor cells mobilized by Angelica polysaccharides (APS).    Methods  The female BALB/c mice irradiated with 8.5 Gy 60Co γ ray were transplanted with peripheral blood mononuclear cells (PBMCs) from male BALB/c mice mobilized by APS. After transplantation, the changes of WBC, HGB and platelet in peripheral blood were observed. Thirty-day survival rate was calculated. PCR was used to verify the cell source of hematopoiesis reconstruction.    Results  The number of WBC, platelet and HGB, and 30-day survival rate of APS group were markedly higher than that of negative control group and normal saline group (P<0.05). PCR results showed Y chromosome sequence existed in the survived mice of APS group, indicating that the repopulated hematopoietic cells came from the male donor.    Conclusion  The hematopoietic stem/progenitor cells mobilized by APS can reconstruct long-term hematopoiesis in mice of hematopoietic aplasia.
