摘要:
目的    观察糖基化牛血清白蛋白(advanced glycated bovine serum albumin, AGE-BSA)对人脐静脉血管内皮细胞(human umbilical vein endothelial cells, HUVEC)小管样结构(tubule like structure,TLS)形成的影响并探讨其可能机制。    方法    实验设实验组、实验对照组和空白对照组。实验组设25、50、100 μg/ml 3个AGE-BSA浓度;同时设置相同浓度的牛血清白蛋白(bovine serum albumin,BSA)作为实验对照组;内皮细胞完全培养基作为空白对照组。各组设24、48、72 h 3个时相点,采用三维胶TLS形成实验和ELISA方法分别检测不同浓度AGE-BSA作用下HUVEC在Ⅰ型胶原中形成TLS的长度以及HUVEC自分泌血管内皮细胞生长因子(vascular endothelial growth factor, VEGF)的浓度。    结果    在24~72 h内,浓度为25~100 μg/ml的AGE-BSA比相同浓度的BSA使HUVEC培养上清中VEGF的量和在胶原中分化形成TLS的长度均明显增加(P<0.05)。    结论    AGE-BSA可通过刺激VEGF自分泌促进内皮细胞TLS形成,提示糖基化终末产物可能参与糖尿病视网膜血管生成。
Abstract:
Objective    To observe the effects of advanced glycated bovine serum albumin (AGE-BSA) on the tubule like structure (TLS) formation of endothelial cells.     Methods    There were three groups: AGE-BSA (25, 50 and 100 μg/ml) treated groups, BSA (25, 50 and 100 μg/ml) treated groups as BSA control groups, endothelial complete medium without AGE-BSA or BSA as blank control groups. After incubation of human umbilical vein endothelial cells (HUVECs) for 24, 48 and 72 h, TLS formation assay and ELISA were used to detect the length of TLS formation on the surface of collagen Ⅰ and the vascular endothelial growth factor (VEGF) autocrine of HUVECs in presence of AGE-BSA.     Results    In 24 to 72 h, AGE-BSA (25, 50, 100 μg/ml) increased the average length of TLS formation on the surface of collagen Ⅰ and the concentration of VEGF autocrine in supernatant of HUVECs(P<0.05).     Conclusion    AGE-BSA drives TLS formation by stimulating VEGF autocrine of HUVECs, suggesting AGEs play an important role in angiogenesis of diabetic retinopathy.
