摘要:
目的     初步探讨ATP对大鼠逼尿肌不稳定膀胱的作用。     方法     建立雌性SD大鼠BOO DI 模型,以正常雌性逼尿肌稳定(detrusor stability, DS)大鼠作对照,制作离体膀胱并Krebs液孵育,膀胱内分别给予0.9%NaCl、ATP、PPADS液后用0.9% NaCl液以0.1 ml/min的速度灌注膀胱,同法灌注膀胱时向浴液中加入ATP以及先浴液中加入PPADS孵育膀胱10 min后再灌注膀胱,并向浴液中加入ATP,记录灌注膀胱和停止灌注后各4 min内的膀胱压力曲线,测量20 s、50 s、4 min、8 min时的膀胱压力值。     结果     离体膀胱内给予NaCl、ATP、PPADS后,2组大鼠膀胱压力曲线形态和组内和组间各时间点膀胱压无显著性差别(P>0.05)。灌注时浴液中加入ATP,离体膀胱出现膀胱收缩,50 s时DS组膀胱压高于BOO DI组;先PPADS孵育膀胱后再加入ATP,膀胱收缩幅度显著降低,50 s时组间膀胱压无差别。     结论     膀胱内给予或膀胱上皮细胞释放的ATP通过神经反射引起尿频和参与逼尿肌不稳定收缩,支配逼尿肌的嘌呤能神经释放的ATP可直接作用于逼尿肌引起收缩。
Abstract:
Objective     To explore the role of adenosine triphosphate (ATP) in the detrusor muscle from unstable bladders of rats.      Methods     There were 30 female Sprague-Dawleys (SD) rats in bladder outlet obstruction (BOO) group and 15 female SD rats in control group. The bladders from 10 rats from each group were incubated in 37 ℃ Kreb solution. After a bolus infusion of sodium chloride, ATP and  pyridoxalphosphate-6-azophenyl-2’4’-disulfonic (PPADS) were infused into the in vitro bladder respectively, and then the bladders were pumped with sodium chloride at 0.1 ml/min for 4 min. A cystometry was performed at the same time for 8 min. Later, ATP was added into the incubation solution with or without incubation of PPADS previously and cystometry was recorded simultaneously. The bladder pressure curves were recorded and the pressure of in vitro bladders at the time points of 20 s, 50 s, 4 min, 8 min were measured.      Results     For the bladder pressure curve and bladder pressure at different time points, there was no significant difference in each group and between the two groups when the drugs were given into bladders. There was a distinctive bladder contraction when ATP was added into the incubation solutions .The bladder pressure of control group was higher than that of BOO group at 50 s (P<0.01). However, the amplitude of bladder contraction was decreased dramatically when the bladder was incubated previously with PPADS. There was no dramatical difference in the bladder pressure between the two groups.      Conclusion     ATP given intravesically or released by the urothelium cells plays a role in the urinary frequency and detrusor instable contraction through nervous reflex. The ATP released by the bladder purinergic nerves can induce detrusor contraction directly.
