摘要:
目的     探讨脂多糖 (lipopolysaccharide, LPS)在体内外对Akt蛋白表达和磷酸化水平的影响。     方法     用不同剂量LPS攻击RAW264.7细胞,检测不同时相点细胞Akt蛋白表达和磷酸化水平。小鼠腹腔注射LPS 6 h 后检测肺组织Akt蛋白表达和磷酸化水平。     结果     不同剂量LPS攻击RAW264.7细胞,细胞Akt蛋白表达没有显著差异(P>0.05),磷酸化水平增加。而内毒素血症小鼠肺组织Akt蛋白表达没有显著变化(P>0.05),磷酸化水平减低。     结论     LPS在体内外对Akt蛋白磷酸化作用不同,体外Akt蛋白磷酸化水平增高,但在内毒素血症小鼠体内出现Akt蛋白磷酸化水平异常减低。
Abstract:
Objective     To explore the effect of LPS on the expression and phosphorylation of Akt, a PTEN/PI3k signal transduction downstream molecule, in vivo and in vitro.      Methods     RAW264.7 cells were attacked with LPS at different concentrations. The Akt protein expression and phosphorylation level were detected by Western blotting at 8, 24, 48 h after attack. Six h after receiving 20 mg/kg LPS intra-peritoneal injection, the mice were killed and their lung tissues were resected for Akt protein expression and phosphorylation.      Results     The Akt protein expression of RAW264.7 cells showed no obvious difference after LPS attack at different doses, but their phosphorylation level was increased obviously with the increase of the doses. Whereas the phosphorylation level of Akt protein was decreased in the lung tissue of mice 6 h after LPS attack.      Conclusion     The phosphorylation level of Akt protein is different after LPS attack in vitro and in vivo. The Akt protein phosphorylation is increased obviously in vitro, but decreased in mice model of endotoxemia.
