摘要:
目的     复制小鼠→大鼠异种心脏移植模型,探讨核转录因子κB (nuclear factor-κB ,NF-κB) P65蛋白在异种移植心肌中的表达及NF-κB的DNA结合活性。     方法     动物随机数字法分成2组:A组(对照组),n=6;B组(移植组),n=6。采用免疫印迹杂交法(Western blot)检测对照组及移植组心肌组织NF-κB P65表达,凝胶电泳迁移率实验检测NF-κB DNA结合活性。     结果     移植组的NF-κB P65蛋白表达和NF-κB DNA结合活性均显著高于对照组(P<0.05)。     结论     NF-κB 的激活可能在大鼠异种移植心脏排斥反应中起着重要作用。
Abstract:
Objective     To reproduce mouse to rat heart transplantation model and determine the expression and DNA binding activity of NF-κB P65 in xenograft heart.      Methods     Six NIH mice were used as heart donors and six Wistar rats as heart recipients. After the transplantation, the transplanted hearts without any drug intervention were prepared for experiment after cardiac arrest. The hearts from another six NIH mice served as normal controls. The expression of NF-κB P65 protein and NF-κB P65 DNA binding activity in xenograft heart were determined by Western blotting and electrophoretic mobility shift assay (EMSA).      Results     The expression level of NF-κB P65 and NF-κB P65 DNA binding activity in transplanted hearts were higher than control hearts (P<0.05).      Conclusion     NF-κB may play an important role in mouse to rat heart transplantation.
