摘要:
目的    观察大鼠血管平滑肌α肌动蛋白基因反义腺病毒载体(pAdanti-α-SMA)转染对转化生长因子β1(TGF-β1)所诱导的肾小管上皮细胞-肌成纤维细胞转分化(TEMT)的影响。    方法    pAdanti-α-SMA载体构建并鉴定,转染肾小管上皮细胞(TEC)后免疫荧光染色和Western blot检测α-SMA水平。实验分为5组,A组:正常肾小管上皮细胞(TEC,对照组);B组:TEC+2 μl  pAdanti-α-SMA;C组:TEC+1   μg天然TGF-β1;D组:TEC+1 μg天然TGF-β1+2 μl pAdanti-α-SMA;E组:TEC+1 μg天然TGF-β1+10 μl pAdanti-α-SMA。    结果    成功构建pAdanti-α-SMA,pAdanti-α-SMA转染TEC后抑制了TGF-β1诱导的TEC 的α-SMA水平并呈剂量依赖性。    结论    pAdanti-α-SMA可有效地抑制肾小管上皮细胞的肌成纤维细胞转分化。
Abstract:
Objective    To evaluate the inhibiting effect of antisense vascular smooth muscle alpha adenovirus vector on tubular epithelial myofibroblast transdifferentiation induced by TGF-β1.      Methods    The pAdanti-α-SMA was constructed and identified. The α-SMA expression in tubular epithelial cells (TEC) was detected by immunofluorescence and Western blot. Five groups were designed as control (A), TEC transfected by 2 μl pAdanti-α-SMA (B), TEC induced by TGF-β1 (C), TEC induced by TGF-β1 and transfected by 2 μl pAdanti-α-SMA (D), TEC induced by TGF-β1 and transfected by 10 μl pAdanti-α-SMA (E).      Results    The pAdanti-α-SMA was successfully constructed. The expression of α-SMA in TEC induced by TGF-β1 was inhibited by pAdanti-α-SMA.       Conclusion    Tubular epithelial myofibroblast transdifferentiation can be inhibited by pAdanti-α-SMA.
