摘要:
目的    探讨茶多酚(tea polyphenols, TP)对缺氧大鼠红细胞增多症的预防作用。    方法    雄性Wistar大鼠,分为平原组、慢性缺氧组、高剂量TP(20 mg/kg)+慢性缺氧组、低剂量TP(10 mg/kg)+慢性缺氧组。大鼠在5 000 m高原环境模拟舱中,间断缺氧(12 h/d), 每2天1次腹腔注射CoCl2溶液(5.5 mg/kg)以建立红细胞增多症模型。经颈动脉插管取血,然后测定大鼠红细胞数、血红蛋白和红细胞压积,并进行血流变学及动脉血气分析。    结果    ①红细胞增多症模型组大鼠的红细胞数、血红蛋白和红细胞压积显著增加,全血黏度等血流变学指标显著上升;②与模型组相比,慢性缺氧大鼠的红细胞数、血红蛋白和红细胞压积值均明显下降,且高剂量TP组较低剂量组显著,但仍高于平原组;同时TP可以改善缺氧大鼠血流变学指标,降低全血黏度及红细胞刚性指数等;但TP对缺氧大鼠的血氧饱和度等血气值没有影响。    结论    TP对缺氧大鼠红细胞增多症有一定预防作用,提示TP对高原红细胞增多症可能有类似作用。
Abstract:
Objective    To observe the protective effects of tea polyphenols (TP) on chronical hypoxia induced polycythemia in rats.     Methods    Wistar rats were divded into 4 groups:(1) the normoxia group, (2) chronical hypoxia group, (3) low dose TP (10 mg/kg)group, (4) high dose TP (20 mg/kg) group. To establish the model of polycythemia, rats in (2), (3) and (4) were exposed to intermittent hypoxia, at simulated altitude of 5 000 m 12 h/d for 40 days, and were given CoCl2 5.5 mg/kg intraperitoneal administration every other day. At the end of the experiment, RBC, HG, HCT and the value of the blood rheology were detected, and arterial blood gas analysis were also done by the analysator.     Results    The value of RBC, HB and HCT in TP groups were all decreased significantly, and the effects of 20 mg/kg TP was more significant than that of 10 mg/kg Furthermore, TP also decreased the whole blood viscosity and RBC rigidity index in chronical hypoxia rats, which indicated that TP is good to the the blood rheology in chronical hypoxia rats. However, it wasn’t significant that the effects of TP on the value of arterial blood gas such as SaO2 in chronical hypoxia rats(P>0.05).     Conclusion    TP may have protective effects on chronical hypoxia induced polycythemia in rats.
