摘要:
目的  探讨胰岛素样生长因子-1对大鼠坐骨神经再生早期的影响。  方法  建立大鼠坐骨神经切割伤模型,通过硅胶管进行桥接,实验组于再生室内注入胰岛素样生长因子-1,对照组注入等量生理盐水,于术后3、5、7、9、11 d取材,用SABC免疫组化法检测轴突再生情况以及雪旺细胞数目、形态和排列方式的变化。  结果  ①雪旺细胞:在对照组,术后3 d,断端间隙内仅可见少量雪旺细胞,术后7 d,雪旺细胞数量明显增多,术后11 d,整个间隙内的雪旺细胞排列呈有序的条索状。在实验组,术后3 d,雪旺细胞即明显增多,以后各相同时相点雪旺细胞数目均明显高于对照组。②再生轴突:在对照组,术后5 d,轴突开始长出,术后7 d轴突快速增长,至第11天,再生的轴突已长过5 mm的间隙到达神经远端。在实验组,术后3 d轴突即开始长出,9 d就快到达远端。  结论  胰岛素样生长因子-1能促进轴突延伸和雪旺细胞增殖,明显促进坐骨神经再生。
Abstract:
Objective    To investigate the role of insulin-like grow factor 1 (IGF-1) on the regeneration of peripheral nerve after operation.      Methods    The right sciatic nerve of adult female SD rats was transected and bridged with silicone tube. IGF-1 was injected into the silicone tube in the treated group, while normal saline instead in control group, with 30 rats in each group. On postoperative day 3, 5, 7, 9 and 11, the regeneration of axons was observed by SABC immunohistochemical methods with anti-neurofilment 200, and the proliferation of Schwann cells was measured with anti-S-100.     Results    Significant difference was seen in the length of regenerating neurites and the number of Schwann cells at all time points between two groups (P<0.01).     Conclusion    Local application of IGF-1 in the regenerative chamber after transaction promotes the proliferation of Schwann cells and the growth of neurites.
