摘要:
目的     分析细胞因子IL-2和IL-10对Treg细胞体外扩增的促进作用,比较扩增的Treg细胞和自然分选Treg细胞对CTL功能活性的影响。     方法     采用不同浓度细胞因子IL-2和IL-10作为诱导剂,分析2种细胞因子体外诱导Treg细胞增殖的有效性及最适浓度,同时以2种细胞因子诱导扩增的Treg细胞和自然分选的Treg细胞分别作用于CTL,分析采用体外扩增Treg细胞抑制CTL活性的可能性。     结果     100 ng/ml IL-2能够诱导Treg细胞增殖明显增加,经48、96 h和144 h诱导后,其增殖比例分别达到10%、18%和20%,而IL-10的使用浓度需达到150 ng/ml才能获得同样的扩增比例;与自然分选的Treg细胞对比,采用细胞因子体外扩增的Treg细胞具有同等抑制CTL功能活性的效力,组间无显著性差异(P>0.05)。     结论     采用添加IL-2培养体系是体外扩增Treg细胞的优选方法,其增殖效能强于IL-10;体外扩增Treg细胞与自然分选Treg细胞对CTL的功能抑制没有明显差异。
Abstract:
Objective     To explore the promoting effects of IL-2 and IL-10 on Treg proliferation in vitro, and to compare the inhibiting effects of the proliferated Treg on CTL with those of the enriched Treg.      Methods     After IL-2 or IL-10 was added into the culture system to stimulate Treg proliferation, the Treg cells were counted at 48, 96 and 144 h to determine the cytokines’ efficacious doses in vitro. After collecting the proliferated Treg, we compared the inhibiting effects of the proliferated Treg on CTL with those of the enriched Treg.      Results     IL-2 and IL-10 induced Treg proliferation in vitro significantly. IL-2(100 ng/ml) increased the number of Treg cells by 10%, 18% and 20% in vitro at 48, 96 and 144 h after stimulation. IL-10 (150 ng/ml) provided the same inducing effects as IL-2, proving less capable. The proliferated Treg had the same efficacy in inhibiting CTL functions as the enriched Treg (P>0.05).      Conclusion     IL-2 is superior to IL-10 for Treg proliferation in vitro. The proliferated Treg is as efficacious as MACS enriched Treg in inhibiting CTL function.
