摘要:

目的 建立小鼠铜绿假单胞菌(Pseudomonas aeruginosa, PA)肺部感染模型,并对该模型进行初步评价。方法 采用一种快速无创的小鼠气管插管滴注法,将小鼠在透射灯照射下经口咽气管插管,滴注1×108 CFU/mL的铜绿假单胞菌悬液50 μL,待接种感染7 d后处死,进行肺部菌落计数和病理学评价。结果 经气管接种感染后,与对照组小鼠比较,感染组小鼠生理状态不佳,肺组织匀浆检出PA,血液中白细胞和中性粒细胞数目显著增多(P<0.05),且出现肺部病理学改变,显示有炎症反应;对照组小鼠肺组织匀浆则未见PA生长,且血液中白细胞和中性粒细胞数目正常,未出现炎症反应。结论 采用小鼠气管插管滴注法,通过呼吸道感染途径可成功建立小鼠肺部PA感染模型,该模型具有稳定性好,易定量的优点

Abstract:

Objective To establish and validate a mouse model of pulmonary infection of Pseudomonas aeruginosa.  Methods The models of pulmonary Pseudomonas aeruginosa infection was established in mice by intratracheal instillation of 50 μL (1×108 CFU/mL) Pseudomonas aeruginosa suspension delivered through orotracheal intubation under a transillumination lamp. At 7 d after the infection, the mice were sacrificed for counting the bacterial colonies in the lungs and for pathological evaluation. Results After intratracheal instillation of  Pseudomonas aeruginosa, the mice showed a poor general condition and had positive findings of Pseudomonas aeruginosa in the lung tissues with significantly increased blood white blood cell and neutrophil counts compared with the control mice (P<0.05); pathological examination revealed obvious pathological changes and inflammations in the lungs of the mice after the infection. In the control mice, no bacteria or pathological changes were detected in the lung tissue, and the white blood cell and neutrophil counts were normal without evidence of inflammatory responses. Conclusion We successfully established a mouse model of pulmonary Pseudomonas aeruginosa infection by delivering the pathogens through tracheal intubation. This model shows a good stability and allows convenient quantitative analysis.
