摘要:

目的       探讨肝X受体β(liver X receptor β,LXRβ)在皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,cSCC)中的表达及其意义。方法       采用PCR array、Western blot检测LXRs在角质形成细胞(keratinocyte,KC)和皮肤鳞状细胞癌A431细胞系中的表达,并采用免疫组化Envision法检测LXRβ在52例日光性角化病(actinic keratosis,AK)、40例鲍温病(Bowen’s disease,BD)、60例cSCC中的表达,分析LXRβ表达与患者临床参数之间的关系。结果       与KC比较,A431细胞中LXRα、LXRβ的mRNA和蛋白水平均升高,以LXRβ升高尤为显著。免疫组化染色结果发现LXRβ在AK、BD、cSCC中表达逐渐升高,以cSCC升高最明显,其表达与肿瘤分化程度、瘤体大小、浸润深度相关(P<0.05),与年龄、性别、发病部位无关(P>0.05)。结论       LXRβ在鳞状细胞癌细胞和组织中表达均升高,且与肿瘤的增殖、分化相关。

Abstract:

Objective       To investigate the expression and significance of liver X receptor β (LXRβ) in cutaneous squamous cell carcinoma (cSCC). Methods       PCR array and Western blotting were performed to detect the expression level of LXRβ transcriptionally and translationally in human normal keratinocytes (KC cells) and skin squamous cell carcinoma cell line A431. Immunohistochemical staining (Envision method) was used to reveal the expression pattern of LXRβ in the tissues from 52 cases of actinic keratosis (AK), 40 cases of Bowen’s disease (BD) and 60 cases of cSCC. The correlation of the expression of LXRβ was analyzed with clinical parameters.  Results       Compared with the KC cells, the mRNA and protein levels of LXRα and LXRβ were higher in the A431 cells, especially those of LXRβ. Immunohistochemical staining demonstrated that LXRβ expression level was increased gradually form AK, BD to cSCC tissues. Its expression was correlated with differentiation, tumor size and invasion (P<0.05), but not with age, gender and tumor location (P>0.05). Conclusion       LXRβ is highly expressed in cSCC cells and tissues, and correlated with cancer proliferation and differentiation.
