摘要:

目的研究七斛血糖制剂对KKAy自发性2型糖尿病小鼠血糖和并发症的影响。方法10只6周龄雌性C57BL/6小鼠(体质量18~20 g)作正常对照组;50只6周龄雌性KKAy小鼠(体质量28~30 g)随机分成5组(n=10),分别为模型对照组(0.3%的羧甲基纤维素钠),七斛血糖制剂低、中、高剂量组(0.75、1.5、3.0 g/kg),阳性对照组(二甲双胍,0.13 g/kg),灌胃给药28 d。于给药第8、15、22、29天测量小鼠随机血糖、体质量及摄食量,于第29天取血检测血脂二项,取肝脏、胰腺组织观察病理改变。结果与C57BL/6正常对照组比较,KKAy模型对照组小鼠摄食量、体质量、随机血糖、总胆固醇和甘油三酯水平显著升高(P<001),小鼠肝细胞肿胀,脂肪沉积明显,胰岛萎缩。在给药第29天,与模型对照组的随机血糖值(26.95±4.62)mmol/L比较,七斛血糖制剂低[(18.47±8.41)mmol/L,P<005]、中[(17.93±5.90)mmol/L,P<005]、高[(15.10±2.60)mmol/L,P<001]剂量组及二甲双胍组[(17.58±7.72) mmol/L,P<001]小鼠的随机血糖值均显著降低,其中七斛血糖制剂高剂量组降糖率达43%。与模型对照组比较,给药15 d后,七斛低、中、高剂量组及二甲双胍组小鼠摄食量和体质量均显著降低(P<005);七斛血糖制剂低、中、高剂量组小鼠的总胆固醇和甘油三酯水平均有降低趋势;七斛血糖制剂中、高剂量组及二甲双胍组小鼠肝细胞肿胀程度、脂肪沉积及胰岛萎缩程度均减轻。结论七斛血糖制剂能够有效降低KKAy小鼠的血糖值,改善血脂水平和减轻肝脏、胰腺的病理改变。

Abstract:

ObjectiveTo assess the effect of the Qihu preparation, a compound preparation of traditional Chinese herbal medicine for treatment of diabetes, on blood glucose levels and organ injures in spontaneous type 2 diabetic KKAy mice. MethodsWith 10 female C57BL/6 mice (6 weeks old) as the normal control group, 50 female KKAy mice (6 weeks old) were randomized equally into 5 groups, namely the model control group, low, moderate and highdose Qihu treatment groups (treated with 0.75, 1.5, 30 g/kg of the crude drugs, respectively), and the positive control group (treated with 0.13 g/kg metformin). The drugs for treatment were administered by gavage twice daily for 28 consecutive days, and the mice in the normal control and model control groups were given an equal volume of 0.3% sodium carboxymethylcellulose. Random blood glucose levels, body weight and food intake of the mice were measured on days 8, 15, 22, and 29 during the treatment. On day 29, orbital blood samples were collected for measurement of plasma levels of total cholesterol (TC) and triglyceride (TG), and the liver and pancreas were collected for examination of pathological changes. ResultsCompared with the normal control mice, KKAy model mice had more food intake, heavier body weight, and higher levels of blood glucose, TC and TG with more obvious liver and pancreas injures. Food intake and body weight were significantly reduced in the mice treated with Qihu and metformin as compared with those in the model control mice. In the mice treated with low, moderate and highdose Qihu preparation and with metformin, the random blood glucose levels at the end of treatments were 18.47±8.41 mmol/L (P<005), 17.93±5.90 mmol/L (P<005), 15.10±2.60 mmol/L (P<001), and 17.58±7.72 mmol/L (P<001), respectively, all significantly lower than that of 26.95±4.62 mmol/L in the model control group, and their levels of TG and TC were both moderately lowered. Pathological examination revealed lessened liver damage and pancreatic atrophy in all the Qihu groups and in metformin group compared with the model control group without significant differences among the 4 drugtreated groups. ConclusionQihu preparation for treatment of diabetes can significantly reduce blood glucose level and alleviate liver and pancreatic injures in KKAy mice.
