摘要:

目的      探讨初生至成年时期大鼠膀胱Cajal间质细胞(interstitial cells of Cajal,ICC)增殖变化情况及其与相应时间点膀胱兴奋性的关系。方法      40只刚出生的大鼠分为2组,每组20只。第1组从出生后第1、15、30、45、60天对其膀胱进行c-kit和Ki67染色,观察ICC细胞的增殖情况;第2组在上述同一时间对其膀胱进行c-kit和SM-actin染色,共聚焦显微镜下观察ICC的分布密度,并测定同期膀胱肌条的兴奋性。结果      膀胱ICC细胞的增殖速度和分布密度从第1天开始逐步增加,到第30天后增殖速度明显下降,分布密度也随之趋于稳定。大鼠逼尿肌肌条兴奋性从出生后第1天逐渐下降,第 45~60天趋于稳定。结论      大鼠膀胱ICC细胞在出生后迅速增殖,到第45天分布密度趋于稳定。膀胱兴奋性随着ICC分布密度的增加逐渐降低,并趋于稳定。

Abstract:

Objective      To observe the proliferation pattern of interstitial cells of Cajal (ICC) in the bladder of rats from birth to adulthood and explore its relationship with bladder excitability. Methods      Forty newborn rats were divided into 2 groups, with 20 rats in each group. The bladder from the first group underwent ckit and Ki67 staining on days 1, 15, 30, 45 and 60 after birth to observe cell proliferation. The bladder from the second group underwent c-kit and SM-actin staining to observe distribution density of ICCs under a confocal microscope, and the excitability of bladder muscle strips was measured at the same time points. Results      The proliferation and distribution density of ICCs in the bladder started to increase since day 1 and culminated on day 30. Then the proliferation significantly slowed down, and the distribution density tended to be stable. The excitability of bladder muscle stripes was in a decrease trend from day 1, and then turned to stable from day 45 to day 60. Conclusion      ICCs in the rat bladder show rapid proliferation since birth, and the distribution density becomes stable on day 45. The excitability of bladder is decreased gradually along with the distribution density increase of the ICCs, and then tends to be stable.
