摘要:

目的      研究第10号染色体缺失的磷酸酶及张力蛋白同源基因蛋白(phosphatase and tensin homolog deleted on chromosome ten, PTEN)在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达及其临床意义。方法      收集30例OSCC及30例距癌组织边界>2 cm的癌旁组织,Western blot测定上述组织PTEN蛋白含量,qRT-PCR测定mRNA的表达水平,免疫组化(immunohistochemistry,IHC)定性测定PTEN蛋白在上述组织细胞中的组织学定位与表达情况,并根据PTEN表达结果分析其与OSCC临床病理关系。结果      ①癌旁组织均为PTEN蛋白阳性表达,PTEN蛋白阳性表达主要为细胞浆内棕黄色或棕褐色染色。OSCC组织多为阴性表达。②OSCC癌组织中PTEN(蛋白和mRNA)的表达明显低于2 cm外的癌旁组织(P<0.05)。③不同年龄、不同性别、不同发生部位OSCC的PTEN(蛋白和mRNA)表达差异无统计学意义(P>0.05)。④PTEN在高分化鳞癌中的表达明显较中低分化鳞癌高(P<0.05)。⑤无淋巴转移的较有淋巴转移的表达高(P<0.05)。⑥临床Ⅰ+Ⅱ期表达明显高于Ⅲ+Ⅳ期(P<0.05)。结论     PTEN在OSCC癌组织中存在不同程度的异常表达,可能是口腔鳞癌发生、发展、侵袭转移过程中的一个重要因素。

Abstract:

Objective      To investigate the expression and clinical significance of phosphatase and tensin homolog deleted on chromosome ten (PTEN) in oral squamous cell carcinoma (OSCC). Methods      Tissue samples from 30 cases of OSCC and from the matched paracarcinoma regions (from 2 cm of the tumors) were collected. Western blot analysis and qRT-PCR were used to detect the expression of PTEN at protein and mRNA levels. And immunohistochemical assay was employed to observed the location and distribution of the protein. The obtained results were analyzed for their relationship with OSCC clinical pathology. Results      ① PTEN protein was positively expressed in the paracancerous tissues, and mainly located in the cytoplasm within the brown or brown stain. But it was not detected in the most OSCC tissues. ② The protein and mRNA levels of PTEN were significantly lower in the OSCC tissues than in the paracancerous regions (P<0.05). ③ There were no significant differences in the protein and mRNA levels of PTEN from different positions of the tumors and among the patients from different age and sex (P>0.05). ④The expression of PTEN were higher in well-differentiated tumors than moderately differentiated and undifferentiated tissues (P<0.05). ⑤The patients without lymph metastasis exhibited significantly increased expression of PTEN compared with those with lymph metastasis (P<0.05). ⑥ PTEN expression levels were significantly higher in the patients of stage Ⅰ+Ⅱ than those of stage Ⅲ+Ⅳ (P<0.05). Conclusion      PTEN is abnormally expressed in the OSCC tissues to some extent, and it may be a key factor in tumorigenesis, invasion and migration of OSCC.
