摘要:

目的      探讨人端粒酶逆转录酶(human telomerase reverse enzyme, hTERT)调控Gli1的表达及其对胃癌细胞侵袭迁移的影响。      方法          采用pIRES2载体构建hTERT过表达质粒,qRT-PCR和Western blot检测hTERT对Gli1表达的影响;构建Gli1启动子荧光素酶质粒,检测hTERT对Gli1启动子荧光素酶活性的影响;构建Gli1干扰质粒,并与hTERT过表达质粒共转染SGC-7901细胞;Transwell实验和划痕实验检测胃癌细胞的侵袭、迁移能力的变化;      结果      过表达hTERT显著上调Gli1 mRNA和蛋白的表达(P<0.01),并显著增加Gli1启动子荧光素酶的活性(P<0.01);而si-Gli1显著抑制Gli1的表达;干扰Gli1的表达,减弱hTERT对胃癌细胞侵袭、迁移能力的影响。      结论      hTERT通过促进Gli1的转录,增加Gli1的表达,进而增强胃癌细胞的侵袭迁移。

Abstract:

Objective      To investigate the mechanism by which human telomerase reverse enzyme (hTERT) regulates Gli1 expression to promote metastasis of gastric cancer cells.       Methods       After down-regulation of Gli1 expression with a specific siRNA against Gli1, the migration and invasion of gastric cancer cells were detected by wound healing and Transwell assays. The protein expression of hTERT and Gli1 was detected by Western blotting. Ectopic expression of hTERT was performed using pIRES2 vector. After the dual luciferase vector that contains the Gli1 promoter was co-transfected into SGC-7901 cells, and the luciferase activity in the cells was measured.       Results      Si-Gli1 markedly inhibited the Gli1 expression, and the down-regulation of Gli1 significantly suppressed the migration and invasion of gastric cancer cells. Ectopic expression of hTERT significantly enhanced the expression of Gli1 at both mRNA and protein levels (P<0.01). Over-expression of hTERT also significantly enhanced the luciferase activity of Gli1 promoter (P<0.01).       Conclusion      hTERT may promote the migration and invasion of gastric cancer cells through enhancing Gli1 expression at transcriptional level.
