摘要:
目的      检测胚胎期小鼠Tbx18 在mRNA及蛋白水平的表达情况,了解Tbx18在小鼠胚胎发育过程中的时空表达模式。      方法      用整体原位杂交技术检测小鼠整胚TBX18 mRNA的表达,用 X-gal染色检测Tbx18-Cre/Rosa26R/LacZ双杂合基因谱系示踪模型小鼠胚胎的报告基因LacZ蛋白的表达,用荧光定量PCR检测小鼠胚胎期心脏mRNA的定量表达。      结果      发现在小鼠胚胎期9.5~10.5 d转录因子Tbx18主要在前体心外膜表达,由前体心外膜逐渐迁移定位于心外膜;在11.5~12.5 d的小鼠胚胎Tbx18主要定位于心脏、上下肢、体节及上下唇鄂区域;大于12.5 d的小鼠胚胎Tbx18除了上述部位外还定位于肾、输尿管、膀胱等区域。      结论      转录因子Tbx18在小鼠胚胎发育过程中的表达具有时空特异性,对小鼠心脏的发育可能起着至关重要的作用;同时在上下肢、体节、唇鄂、肾、输尿管、膀胱的发育中也可能起非常重要的作用。
Abstract:
Objective      To detected the mRNA and protein expression of transcription factor Tbx18, and to reveal its spatiotemporal expression during mouse embryonic development.       Methods      The Tbx18 mRNA expression in whole embryo during mouse embryonic development was detected by whole mount in situ hybridization. The protein expression of reporter LacZ gene in Tbx18-Cre/Rosa26R/LacZ genetic lineage tracer model of mouse embryos was detected using X-gal staining. The mRNA expression Tbx18 in mouse embryonic heart were detected using fluorescence quantitative PCR.       Results      The Tbx18 signals of the E9.5 d to E10.5 d mouse embryos were mainly expressed in the proepicardium at first and then emigrated to the epicardium gradually. The Tbx18 signals of the E11.5 d to E12.5 d mouse embryos were mainly expressed in epicardium, forelimb bud and hindlimb bud, somite and mandibula/maxillar cleft region. Besides the regions above, the Tbx18 signals of the mouse embryos older than E12.5 d were also expressed in the kidney, ureter and bladder.       Conclusion      The expression of the transcription factor Tbx18 has spatiotemporal idiosyncrasy in mouse embryonic development. It may play a vital role in mouse heart development, and also in the development of kidney, ureter, bladder, somite, forelimb and hindlimb buds, and mandibular/maxillar cleft region.
