摘要:
目的      探讨外源性硫化氢对四氯化碳(CCl4)诱导大鼠肝纤维化的预防作用。      方法      将24只Wistar雄性大鼠随机分为3组,每组8只,即正常对照组、模型组、硫化氢(H2S)预防组。H2S预防组除腹腔注射40% CCl4外,还给予NaHS腹腔注射,1次/d,至第8周,所有大鼠均被处死,收集大鼠血液,检测肝纤维化指标;取肝组织检测脂质过氧化指标及观察肝脏病理变化;应用RT-PCR和Western blot方法检测肝组织NF-κB的表达。      结果      外源性H2S能降低血清HA、LN、PCⅢ及肝组织MDA含量(P<0.01或P<0.05),增加肝组织SOD、GSH-Px活性(P<0.01),减轻肝纤维化程度(P<0.05),而且能使NF-κB表达明显减少。      结论      外源性硫化氢对CCl4诱导的大鼠肝纤维化具有保护作用,其机制可能与硫化氢抑制NF-κB表达有关。
Abstract:
Objective      To study the preventive effect of exogenous hydrogen sulfide (H2S) on carbon tetrachloride(CCl4)-induced hepatic fibrosis in rats.       Methods       Twenty-four male Wistar rats were randomly divided into normal control group (n=8), model group (n=8), and H2S prevention group (n=8). NaHS was injected into the abdominal cavity of rats in H2S prevention group in addition to 40%CCl4, once a day, for 8 weeks. Then, all rats were sacrificed and blood samples were collected to test the indications of hepatic fibrosis. Lipid peroxide indications and liver pathological changes were observed in liver tissue samples. Expression of NF-κB in liver tissue was detected by RT-PCR and Western blotting, respectively.       Results      Exogenous H2S significantly decreased the serum HA,LN,PCⅢ I and MDA levels, hepatic fibrosis, expression of NF-kB and increased the SOD and GSH-Px activities in liver tissues (P<0.01 or P<0.05).       Conclusion      Exogenous H2S protects against CCl4-induced hepatic fibrosis in rats by inhibiting the expression of NF-κB.
