摘要:
目的      探讨蛋白激酶C epsilon(protein kinase C epsilon,PKC-ε)及碱性调宁蛋白(h1 calponin ,h1 CaP)在妊娠小鼠子宫平滑肌细胞中总蛋白和磷酸化蛋白的时空表达变化及与足月临产、早产的关系和机制。      方法      建立小鼠酒精灌胃早产模型。按妊娠阶段的不同,将小鼠分为未孕组、早孕组(孕7 d)、中孕组(孕12 d)、晚孕组(孕18 d)、足月产组和早产组,每组各10例。应用Western blot检测子宫平滑肌中PKC-ε、磷酸化PKC-ε(pPKC-ε)、h1 CaP、磷酸化h1 CaP(ph1 CaP)的蛋白表达。      结果       足月产组和早产组PKC-ε蛋白表达均显著高于未孕、早孕、中孕组(P<0.01),晚孕组、足月产组、早产组相比较差异无统计学意义(P>0.05);足月产组和早产组h1 CaP蛋白表达均显著高于未孕、早孕、中孕组(P<0.01),晚孕组、足月产组、早产组相比较,差异无统计学意义(P>0.05);足月产组和早产组pPKC-ε/PKC-ε值均显著高于未孕、早孕、中孕组及晚孕组(P<0.01),足月产组与早产组相比较差异无统计学意义(P>0.05);足月产组和早产组ph1 CaP/h1 CaP值均显著高于未孕、早孕、中孕组及晚孕组(P<0.01),足月产组与早产组相比较差异无统计学意义(P>0.05);pPKC-ε/PKC-ε的变化与ph1 CaP/h1 CaP的变化呈正相关(r=0.73,P<0.05)。       结论       PKC-ε及h1 CaP蛋白表达在临产前即达到最高峰,pPKC-ε及ph1 CaP水平在临产时达最高峰。PKC-ε可通过激活h1 CaP参与足月产及早产的分娩发动。
Abstract:
Objective       To evaluate level changes of protein kinase C epsilon (PKC-ε)/phosphorylated PKC-ε (pPKC-ε) and h1 calponin (h1 CaP)/phosphorylated h1 CaP (ph1 CaP) in myometrium of pregnant mice during pregnancy, and to investigate their relationship with labor at term and preterm and the involved mechanisms.       Methods       Pregnant mice were given 6 mg/g ethanol by gavage to construct mouse model of preterm labor. According to different periods of pregnancy, mice were divided into a non-pregnancy(NP) group, an early pregnancy (EP, gestational day 7) group, a middle pregnancy (MP, gestational day 12) group, a late pregnancy (LP, gestational day 18) group, a term labor (TL) group and a preterm labor (PL) group. Each group included 10 mice. Protein levels of PKC-ε/pPKC-ε and h1 CaP/ph1 CaP in mice myometrium were detected by Western blotting.       Results       Protein levels of PKC-ε and h1 Cap in the TL and PL groups were significantly higher than those in the NP, EP and MP groups (P<0.01), while the differences among the LP, TL and PL groups were not significant (P>0.05). The ratios of pPKC-ε/PKC-ε and ph1 CaP/h1 CaP in the TL and PL groups were significantly higher than those in the NP, EP, MP and LP groups (P<0.01), while the difference between the TL and PL groups was not significant (P>0.05). The change of pPKC-ε/PKC-ε was positively correlated with the change of ph1 CaP/h1 CaP (r=0.73, P<0.05).       Conclusion       Protein levels of PKC-ε and h1 CaP in mice myometrium both reach the peak at pre-labor. Levels of pPKC-ε and ph1CaP both reach the peak at parturition. PKC-ε is suggested to contribute to the initiation of term and preterm labor through h1 CaP phosphorylation.
