摘要:
目的      研究脂多糖(lipopolysaccharide,LPS)诱导RAW 264.7细胞促炎性细胞因子IL-1β、IL-6及抗炎性细胞因子IL-10的动态变化规律及八肽胆囊收缩素(cholecystokinin octapeptide,CCK-8)对其表达的影响。      方法      LPS诱导RAW 264.7细胞不同时间(0、3、6、12、24、48 h),在各细胞因子表达高峰的时间点用ELISA及RT-PCR法检测不同剂量(10-10、10-9、10-8、10-7、10-6 mol/L)CCK-8对LPS诱导RAW 264.7细胞IL-1β、IL-6、IL-10蛋白及mRNA表达的影响。      结果      ①LPS可诱导RAW 264.7细胞IL-1β、IL-6、IL-10蛋白合成及mRNA表达增加(P<0.05,P<0.01):各细胞因子蛋白的浓度分别于LPS刺激后6、24、24 h达到高峰[分别为(209.41±10.04)、(2 071.64±9.08)、(207.65±16.27)ng/L];各细胞因子mRNA的表达分别于LPS刺激后3、24、24 h达到高峰[分别为(0.723±0.030)、(1.157±0.045)、(0.767±0.035)]。②CCK-8可剂量依赖性抑制IL-1β、IL-6的表达,并使IL-10表达进一步增加(P<0.01),且孵育细胞24 h,CCK-8上调LPS诱导IL-10生成的幅度要大于孵育48 h。      结论      CCK-8通过抑制LPS诱导RAW 264.7细胞抑制促炎性细胞因子IL-1β、IL-6的表达和进一步增加抗炎性细胞因子IL-10的表达参与抗炎反应过程。
Abstract:
Objective      To observe the dynamic changes of proinflammatory cytokines IL-1β, IL-6 and anti-inflammatory cytokines IL-10 in LPS-induced RAW 264.7 cells and observe the effects of cholecystokinin octapeptide (CCK-8) on their expression.       Methods      After RAW 264.7 cells were stimulated with 1 mg/L LPS for 0, 3, 6, 12, 24 and 48 h respectively, the expression peak times of IL-1β, IL-6 and IL-10 were selected to investigate the effects of CCK-8 at the doses of 10-10, 10-9, 10-8, 10-7 and 10-6 mol/L respectively. Cytokines protein and mRNA expressions were assayed by ELISA and RT-PCR.       Results      The expression of IL-1β, IL-6 and IL-10 mRNA and protein were induced by LPS in RAW 264.7 cells in a time-dependent manner. CCK-8 inhibited LPS-induced IL-1β and IL-6 expression and augmented IL-10 expression in a dose-dependent manner. When the cells were treated for 24 h, CCK-8 augmentation resulted in higher LPS-induced IL-10 expression when compared with that after 48 hours’ treatment.       Conclusion      CCK-8 may participate in the anti-inflammatory response by inhibiting the expression of proinflammatory cytokines IL-1β and IL-6, meanwhile augmenting the expression of anti-inflammatory cytokine IL-10 induced by LPS.
