摘要:
目的      研究沉默核糖核酸酶抑制因子(ribonuclease inhibitor,RI)基因表达,对人膀胱癌BIU-87细胞增殖和侵袭能力及肿瘤生长的影响。      方法      通过用RI mRNA的特异性siRNA表达载体和无同源性的对照载体,在脂质体介导下稳定转染膀胱癌BIU-87细胞,通过筛选、鉴定后,用Am-blue法检测细胞增殖能力及用Western blot分析细胞中MMP-2和MMP-9的表达。将siRNA RI BIU-87(实验组)及对照组细胞皮下注射到BALB/c裸鼠中,观察肿瘤的生长和肿瘤微血管密度的变化,以及nm23-H1和 E-cadherin在肿瘤中的表达。      结果      RI基因表达下调显著增加了细胞的增殖,而且显著增强了MMP-2和MMP-9的表达。动物实验结果显示,实验组显著促进了肿瘤的生长(促瘤增长率31.85%),具有更大的瘤质量和更高的肿瘤微血管密度以及更低的nm23-H1和 E-cadherin的表达。      结论      抑制RI基因的表达能显著提高膀胱癌细胞的生长和侵袭潜能,RI可能作为治疗膀胱癌的靶蛋白。
Abstract:
Objective      To investigate the effects of down-regulating human ribonuclease inhibitor (RI) on cell proliferation, invasion and tumor growth in human bladder cancer BIU-87 cells.       Methods      Two specific siRNA RI expression vectors and a non-homologous vector negative control were transfected into noninvasive bladder cancer BIU-87 cells, then screened and identified. Cell proliferation was measured by Am-blue assay, and the expression of MMP-2 and MMP-9 were detected by Western blotting. Tumor cells at  2×106, including the BIU-87 siRNA3 cells and the BIU-87 vector cells were respectively injected subcutaneously into the back of BALB/c nude mice to observe tumor growth and microvessel density. Immunohistochemical assays for RI, CD31, nm23-H1 and E-cadherin were performed to detect the effect of RI expressions on angiogenesis and metastasis potential of the tumor mass.       Results      Silencing RI expression in BIU-87 cells obviously enhanced cell proliferation, and increased MMP-2 and MMP-9 expression. BALB/c nude mice receiving an injection of RI siRNA transfected BIU-87 cells showed a significant facilitation of the tumor with heavier tumor weight, higher density of microvessel, lower nm23-H1 and E-Cadherin expression than those in the control group.       Conclusion      Silencing RI promotes growth and metastasis potentials of BIU-87 cells. The results show that RI may be a therapeutic target protein for bladder cancer.
