目的     探讨埃滋蛋白(Ezrin)和细胞间黏附分子1(intercellular adhesion molecules 1,ICAM-1)在结肠癌组织中的表达及意义。     方法       应用免疫组织化学S-P法和Western blot方法检测32例结肠癌及癌旁正常组织中Ezrin和ICAM-1的表达,分析其与结肠癌临床病理特点的关系。     结果       ①Ezrin和ICAM-1在结肠癌组织中高表达,正常组织低表达或不表达。Western blot检测结果:Ezrin在癌组织中表达量为(2 121.0±631.5),在正常组织中为(907.8±258.3);ICAM-1在癌组织中表达量为(2 511.5±627.2),在正常组织中为(676.2±323.6),免疫组化显示二者主要表达于细胞膜和细胞质,肿瘤间质中也可见ICAM-1表达,且癌组织中的表达明显高于正常组织。②二者在结肠癌Dukes C+D期中的表达明显高于A+B期(P<0.01),在有淋巴结转移组的表达明显高于无转移组(P<0.01)。③Ezrin和ICAM-1表达成正相关(r=0.898,P<0.01)。     结论       Ezrin和ICAM-1在结肠癌组织中均有表达,其表达程度与结肠癌的分期、有无淋巴结转移有关。
Abstract:
Objective       To study the expression of Ezrin and intercellular adhesion molecule 1 (ICAM-1) in colon cancer and its clinical significance.      Methods        Expression of Ezrin and ICAM-1 in 32 colon cancer tissue samples and 32 its adjacent normal tissue samples was detected by immunohistochemistry S-P and Western blot, respectively. Its relation with the cliniopathological characteristics of colon cancer was analyzed.      Results       Western blot showed that Ezrin and ICAM-1 were highly expressed in colon cancer tissue samples, lowly or not expressed in normal colon tissue samples (P<0.01) .The expression level of Ezrin was 2 121.0±631.5 in colon cancer tissue samples and 907.8±258.3 in its adjacent normal tissue samples, while the expression level of ICAM-1 was 2 511.5±627.2 in colon cancer tissue samples and 676.2±323.6 in its adjacent normal tissue samples (P<0.01). Immunohistochemistry demonstrated that Ezrin and ICAM-1were mainly expressed in cytomenbrane and cytoplasm. ICAM-1 was also expressed in intercellular substance. The ICAM-1 expression level was significantly higher in colon cancer tissue samples than in its adjacent normal tissue samples(P<0.01). The expression level of Ezrin and ICAM-1 was significantly higher in Dukes C+D colon cancer than in Dukes A+B colon cancer(P<0.01), and in colon cancer with lymphatic metastasis than in that without lymphatic metastasis (P<0.01). The expression of Ezrin and ICAM-1 was positively related(r=0.898,P<0.01).      Conclusion        Ezrin and ICAM-1 are expressed in colon cancer tissue, and their expression levels are related with the sage and lymphatic metastasis of colon cancer.
