摘要:
目的    观察雌激素处理对成年雄性小鼠黑质多巴胺神经元数量的影响及可能的调控机制。    方法      选用雄性C57/BL6小鼠10只,分成对照组、雌激素组(E2),采用免疫组化检测雌激素处理对小鼠黑质多巴胺神经元与calbindin阳性神经元数量的改变;采用RT-PCR分析雌激素处理对小鼠中脑内多巴胺转运蛋白(DAT)、胰岛素样生长因子1受体(IGF-1R)与脑源性神经营养因子(BDNF)mRNA水平的影响。    结果      雌激素处理可以增加小鼠黑质多巴胺神经元数量[E2组(119.7±2.9)个vs对照组(93.5±5.1)个,P=0.004],并增加小鼠中脑内DAT的表达水平(P<0.05),而对calbindin阳性神经元数量无影响[E2组(88.0±2.0)个vs对照组(86.2±1.5)个]。雌激素处理同时可以上调小鼠中脑内IGF-1R与BDNF mRNA水平(P<0.05)。    结论      雌激素通过调节IGF-1R与BDNF的表达增加小鼠中脑内多巴胺神经元数量。
Abstract:
Objective     To determine the effect of estrogens (E2) on dopamine neurons in the substantia nigra (SN) of adult male mice and the underlying molecular mechanism.     Methods     Ten C57/BL6 mice were randomized to 2 groups, control group (with subcutaneous  injection of 0.1 ml sesame oil for 10 d) and E2 group (with subcutaneous injection of 2 μg/d 17-β estradiol solved in 0.1 ml sesame oil for 10 d). Immunohistochemistry was used to detect the positive neurons of tyrosine hydroxylase (TH) and calbindin. The expression levels of dopamine transporters (DAT), insulin-like growth factor 1 receptor (IGF-1Rm) and brain-derived neurotrophic factor (BDNF) were detected by RT-PCR.     Results     E2 increased the number of dopamine neurons [(119.7±2.9) vs (93.5±5.1)] (P=0.004) and the mRNA levels of DAT(P<0.05), but had no effect on the number of calbindin-positive neurons [(88.0±2.0) vs (86.2±1.5) ](P=0.507). E2 increased the mRNA levels of IGF-1R and BDNF in the midbrain significantly(P<0.05).     Conclusion     E2 can increase the number of dopamine neurons through upregulating the expression of IGF-1R and BDNF.
