摘要:
目的    研制一种适合我国国情的人血清角膜中期保存液。    方法    配制人血清浓度分别为10%(A组)、20%(B组)、30%(C组)、0%(D组)的角膜保存液保存兔角膜,用Optisol-GS角膜保存液作对照组,分别在保存第3、7、10、14天进行角膜内皮细胞活性率检测、HE染色、扫描电镜超微结构观察,并检测角膜保存液的理化性质及生物安全性。    结果    兔角膜保存至第14天时,C组角膜内皮细胞活性率与对照组比较差异无明显统计学意义(P>0.05),C组被保存的角膜组织病理学和超微结构与对照组Optisol-GS比较无显著差异。各组血清角膜保存液理化性质稳定,无生物危险性。D、A、B组与对照组比较,角膜内皮细胞活性率分别在保存第3、7、10天时差异具有明显的统计学意义(P<0.05);角膜组织病理学比较,在保存第7天,各实验组与对照组无明显差异,随着时间延长,D、A、B组先后依次出现组织病理学改变;超微结构比较,在保存第7天时,D组与C组及对照组具有明显差异。    结论    30%浓度的人血清角膜保存液具有良好的兔角膜保存效果,其理化性质稳定且安全, 可保存兔角膜7~10 d。
Abstract:
Objective    To develop a medium-term corneal storage medium containing human serum and investigate its preservation effect on rabbits’ corneas.     Methods     Corneal storage media containing human serum 10% (group A), 20% (group B), 30% (group C), and 0% (group D) were prepared. Rabbits’ corneas were stored in the above corneal storage media as well as Optisol-GS cornea storage medium (control group). On the 3rd, 7th, 10th and 14th day after preservation, the viability of corneal endothelial cells, the corneal histopathology, the corneal microstructure, and the physicochemical properties and biological safety of corneal storage media were examined using trypan blue-alizarin red staining, pathological sections of corneas, scanning electron microscopy, and microbiological evaluation, respectively.     Results    On the 14th day after preservation, the viability of corneal endothelial cells, the corneal histopathology and the corneal microstructure showed no significant differences between group C and the control group (P>0.05). Besides, the physicochemical properties of corneal storage media were stable without any biological hazard. On the 3rd, 7th and 10th day after preservation, the viability of the corneal endothelial cells showed significant differences between groups A, B and D and the control group (P<0.05). On the 7th day after preservation, the corneal histopathology showed no significant differences between the experimental groups and the control group, and histopathological changes occurred in groups D, A and B sequentially with time. At the 7th day after preservation, the corneal microstructure showed significant differences between group D and group C or the control group.     Conclusion    The medium-term corneal storage medium containing human serum 30% can preserve rabbits’ corneal tissues for 7 to 10 d. It has stable physicochemical properties, and it is safe and effective.
