目的  探讨CEACAM6、MMP-9在胃癌组织中的表达及其与微血管生成之间的关系。  方法  免疫组化法分别检测CEACAM6、MMP-9在54例胃癌组织及10例胃正常组织中的表达情况及与微血管密度(microvessel density,MVD)表达的相关性。  结果  CEACAM6、MMP-9在胃癌组织中表达率分别为85.19%(46/54)、74.7%(40/54),明显高于胃正常组织中表达率30%(3/10)、20%(2/10),胃癌组织中MVD值(32.17±8.97)明显高于胃正常组织[(18.62±5.07),P<0.05)]。胃癌组织中CEACAM6与性别、年龄、分化程度和淋巴结转移无关(P>0.05);而与浸润深度、临床分期有关(P<0.05)。MMP-9和MVD与性别、年龄无关(P>0.05);而与浸润深度、临床分期、分化程度及淋巴结转移有关(P<0.05);CEACAM6与MMP-9在胃癌组织中的表达呈正相关(r=0.593,P=0.000);CEACAM6、MMP-9阳性表达的MVD明显高于相应阴性表达(P<0.05)。  结论  CEACAM6、MMP-9在胃癌组织中高表达,可能参与了胃癌的发生、发展,与胃癌微血管生成和浸润、转移有关。
Abstract:
Objective  To investigate the expressions of CEACAM6 (CD66c) and MMP-9 in gastric cancer tissue and their relationship with tumor micro-angiogenesis.   Methods  Immunohistochemistry was used to detect the expressions of CEACAM6 and MMP-9, and microvessel density (MVD) in 54 samples of gastric cancer and 10 normal gastric tissue. The clinical data were retrospectively analyzed.   Results  The positive rates of CEACAM6 and MMP-9 in gastric carcinoma tissue were 85.19% (46/54) and 74.7% (40/54), respectively, which were significantly higher than those in normal gastric tissue [30.00% (3/10), 20.00% (2/10), respectively, P<0.05]. The value of MVD in gastric cancer tissue was 32.17±8.97, which was significantly higher than that in normal gastric tissue (18.62±5.07, P<0.05). The expression of CEACAM6 was associated significantly with depth of invasion and clinical stage of gastric cancer (P<0.05), but was not associated with gender, age, differentiation and lymph node metastasis (P>0.05). The expression of MMP-9 and the value of MVD were associated significantly with depth of invasion, clinical stage, differentiation and lymph node metastasis of gastric cancer (P<0.05), but they had no relation with gender and age (P>0.05). There was a significantly positive correlation between CEACAM6 expression and MMP-9 expression in gastric cancer tissue (r=0.593, P=0.000). The value of MVD in the individuals with positive expression of CEACAM6 or MMP-9 was significantly higher than that in the patients negative to CEACAM6 or MMP-9 (P<0.05).   Conclusion  Moderate and strong expressions of MMP-9 and CEACAM6 in gastric cancer tissue are associated with tumor micro-angiogenesis, invasion and metastasis, which may contribute to the occurrence and development of gastric cancer.
