摘要:
目的    验证钙结合酪氨酸磷酸化调节蛋白(calcium-binding tyrosine phosphorylation-regulated protein,CABYR)不同异构体之间寡聚体的形成。    方法      人精子提取物进行非还原和还原条件下的SDS-PAGE、二维IEF/SDS-PAGE、对角线电泳、二维 blue native/SDS-PAGE电泳和免疫印迹分析。    结果      在人精子中丰度最高的CABYR亚型是相对分子质量86×103仅含CRA编码区表达序列,具有钙结合能力的异构体。 同时也存在既含CRA又含CRB编码区表达序列的CABYR亚型。在非还原条件下,丰度最高的130×103二聚体是由仅含CRA表达序列的86×103异构体和既含CRA又含CRB表达序列的50×103异构体组成。(200~220)×103寡聚体是由86×103、50×103异构体,以及35×103 (可能既含CRA又含 CRB表达序列)的异构体和仅含CRA表达序列的30×103异构体组成。     结论      证实CABYR异构体可形成寡聚体,CABYR 主要通过86×103和50×103异构体形成异质二聚体,与其他CABYR异构体结合进一步形成更大的寡聚体,参与了人精子纤维鞘超分子结构的装配和形成。
Abstract:
Objective    To confirm the formation of oligomers by different isomers of CABYR, a calcium-binding tyrosine phosphorylation-regulated protein.     Methods      Different human sperm extracts were analyzed by SDS-PAGE, 2D IEF/SDS-PAGE, 2D diagonal gel electrophoresis, 2D blue native/SDS-PAGE and Western blotting, respectively, in native or non-reducing condition and reducing condition.      Results     The most abundant and predominant subtype of CABYR in human sperms was CABYR-A with a 86×103 variant. Some variants contained both CABYR-A and CABYR-B. CABYR isomers were oligomerized by binding through disulfide bond. A 130×103 heterodimer was composed of a 86×103 CABYR-A variant and a 50×103 variant containing both CABYR-A and CABYR-B. A (200-220)×103 isomer contained 86×103 variant and a 50×103 variant, as well as a 35×103 variant (may containing both CABYR-A and CABYR-B) and a 30×103 variant (CABYR-A only).      Conclusion     CABYR isomers can form oligomers mainly by 86×103 and 50×103 variants, and form large oligomers by binding to other CABYR isomers. CABYR isomers participate in assembly and formation of supra molecular structure of fibrous sheath in human sperms.
