摘要:
目的   研究两性霉素B脂质体(amphotericin B liposome, AmBL)胶联羊膜药膜的体内外缓释性能。   方法   将两性霉素B脂质体与纤维蛋白胶混合,再与羊膜胶联,制备成两性霉素B脂质体胶联羊膜药膜,光镜及扫描电镜观察形态学。将药膜置于生理盐水中,连续10 d置换浸出液,高效液相色谱法测定药物浓度。兔右眼眼表行药膜移植,考察房水中药物浓度变化,左眼滴入自制的两性霉素B脂质体眼液作为对照。   结果   药物-纤维蛋白胶与羊膜基底面贴合紧密,药物附着在胶原纤维网状孔隙内。浸出液中前3 d药物浓度分别为59.9、34.1、27.9 μg/ml,以后逐渐减少,第10天为0.78 μg/ml。兔眼表药膜移植后,前2 d房水中药物浓度较高,分别为113.1、61.2 ng/ml,以后逐渐降低;对照组房水中药物浓度维持在较低水平,平均值为26.1 ng/ml。   结论   两性霉素B脂质体胶联羊膜药膜是一种较好的药物缓释系统。
Abstract:
Objective   To study the slow-release properties of amphotericin B liposome(AmBL)-fibrin glue(FG)-amniotic membrane(AFAM)in vivo and in vitro.    Methods   AmBL was added into FG and cross-linked with AM to prepare the AFAM. Morphology of the AFAM was observed under light microscope and scanning electron microscope (SEM). AFAM was placed in saline which was replaced once a day for 10 d. Concentration of the released AmB was assayed by high performance liquid chromatography (HPLC). AFAM was transplanted to the right eyes of rabbits, and changes of AmB concentration in aqueous humor were observed. AmBL solution was dropped into the left eyes which served as controls.    Results   The AmBL-FG was firmly fixed on the basal surface of amniotic membrane. AmBL was attached to the FG meshes. The concentration of AmB in saline was 59.9, 34.1 μg/ml, and 27.9 μg/ml respectively in the initial 3 days and 0.78 μg/ml on day 10, which was then gradually reduced. After transplantation of AFAM, the AmB concentration in aqueous humor was 113.1 and 61.2 ng/ml respectively in the first 2 days, which was then gradually reduced. The AmB concentration in aqueous humor of the control group was relatively low with a mean level of 26.1 ng/ml.    Conclusion   AFAM is a rather good slow-release system of drugs.
