摘要:
目的  研究常氧与缺氧条件下慢性牙周炎大鼠血清中基质金属蛋白酶2(matrix metalloproteinase-2, MMP-2)的表达变化,探讨MMP-2与高原牙周病的关系。  方法  模拟高原缺氧建立大鼠慢性牙周炎模型,采用ELISA法测定正常对照组、常氧牙周炎组、缺氧对照组、缺氧牙周炎组大鼠血清中MMP-2表达。  结果  正常对照组、常氧牙周炎组、缺氧对照组、缺氧牙周炎组血清中MMP-2浓度分别为(92.00±21.78)、(118.09±25.01)、(102.03±21.30)、(159.19±34.13) ng/ml,常氧牙周炎组与正常对照组、缺氧牙周炎组与缺氧对照组、缺氧对照组与正常对照组、缺氧牙周炎组与常氧牙周炎组比较均有统计学意义(P<0.01)。其中缺氧牙周炎组血清中MMP-2浓度升高最为显著(P<0.01)。  结论  缺氧上调血清中MMP-2的表达,进而促进大鼠牙周组织的破坏。
Abstract:
Objective  To study the change of matrix metalloproteinase-2 (MMP-2) expression in serum of rats with chronic periodontitis under normal oxygen or hypoxia conditions and the correlation between MMP-2 and high altitude periodontitis.   Methods  A chronic periodontitis model of rat was established by simulating high altitude hypoxia. Rats were divided into normal control (NC) group, normal oxygen periodontitis group (P1), hypoxia control group(H), and hypoxia periodontitis group (P2). Expression of MMP-2 in serum of rats was detected by ELISA.   Results  The MMP-2 level in N, P1, H, and P2 groups was 92.00±21.78, 118.09±25.01, 102.03±21.30, and 159.19±34.13 ng/ml, respectively (P<0.01). The serum MMP-2 level was significantly higher in P2 group than in other groups.   Conclusion  Hypoxia up-regulates MMP-2 expression in serum and promotes destruction of periodontal tissue in rats, suggesting that MMP-2 participates in destruction of periodontal tissue during hypoxia periodontitis, which may be related with the recurrence and development of high altitude periodontal diseases.
