摘要:
目的  研究丰富与贫瘠环境对缺氧缺血性脑损伤(hypoxic-ischemic brain damage, HIBD)后脑功能的影响。  方法  制备3 d龄SD大鼠HIBD模型,随机数字表法分为丰富环境组(enriched environment, EE)、贫瘠环境组(impoverished environment, IE)和标准环境组(standard environment, SE)。另设假手术组作为对照。各组大鼠于术后第2天开始分别给予不同环境刺激。术后32 d,应用悬吊和斜坡实验测试感觉运动功能,Morris水迷宫实验了解学习记忆能力,免疫组织化学染色检测皮层和海马胶质纤维酸性蛋白(glial fibrillary acidic protein, GFAP)的表达。  结果  感觉运动和学习记忆能力测试发现,IE组明显差于SE组(P<0.01),SE组差于Sham组和EE组(P<0.01),Sham组和EE组无显著差异(P>0.05)。皮层和海马GFAP免疫组织化学染色结果显示,Sham组GFAP阳性细胞数分别低于其他3组(P<0.01),而SE组低于EE组,高于IE组(P<0.01)。  结论  丰富环境增加新生鼠脑损伤后GFAP的表达,改善脑功能;贫瘠环境加重脑功能的损害。
Abstract:
Objective   To investigate the effects of enriched environment and impoverished environment on brain function after hypoxic-ischemic brain damage (HIBD).    Methods   Three-day-old SD rats, which were divided into enriched environment group (EE), standard environment group (SE) and impoverished environment group (IE) by random digits table were used to establish HIBD model. The sham-operation rats served as control group. Different environment stimulations were administrated to the rats respectively since the 2nd day after HIBD. On the 32nd day, hanging test and incline plane test were carried out to evaluate the sensorimotor function. Morris water maze was used to evaluate the learning and memory ability. The expression of GFAP in the cortex and hippocampus was measured by the method of immunohistochemistry.    Results   The brain function of IE group was much worse than that of SE group (P<0.01), and the function of SE group was worse than that of sham group and EE group(P<0.01). There was no significant difference between sham group and EE group (P>0.05).Immunohistochemical analysis showed the expression of GFAP in the cortex and hippocampus of Sham group was significantly lower than that of other groups (P<0.01), and the expression of GFAP of SE group was lower than that of EE group, but higher than that of IE group (P<0.01).    Conclusion   Enriched environment stimulation increases the expression of GFAP in the brain of premature rats after HIBD, and improves the brain function. Impoverished environment stimulation impairs the brain function.
