摘要:
目的   了解P2X7受体在新生大鼠缺氧缺血型脑损伤后的表达变化。  方法  选择出生7 d的SD大鼠建立新生鼠缺氧缺血性脑损伤模型,通过Western blot和免疫组化检测缺氧缺血前后脑皮层、白质、海马P2X7受体表达。  结果  缺氧缺血后2 h,患侧皮层、白质和海马P2X7受体蛋白较假手术对照组明显下调(P<0.05,P<0.01);免疫组化也显示患侧皮层、白质和海马P2X7受体表达下降。  结论  P2X7受体可能参与了新生鼠缺氧缺血性脑损伤过程。
Abstract:
Objective   To investigate the change of P2X7 receptor expression in neonatal rats after hypoxic-ischemic brain damage (HIBD).    Methods   Twenty SD rats were randomized into 2 groups, control group and hypoxic-ischemic (HI) group. Western blotting and immunohistochemistry was used to detect the protein expression of P2X7 receptor in cerebal cortex, subcortical white matter and hippocampus before and after hypoxia-ischemia.    Results   In comparison to the sham operation controls, a significant down-regulation of the P2X7 receptor protein was observed in ischemic cerebral cortex, subcortical white matter (P<0.05) and hippocampus (P<0.01) of ischemic hemisphere 2 h after hypoxia ischemia. A substantial loss of ipsilateral P2X7R immunostaining in the lateral corpus callosum, adjacent cortex, and hippocampus was observed in comparison to the controls.    Conclusion   The post-ischemic downregulation of P2X7R indicates a role for this receptor in the pathophysiology of hypoxic-ischemic brain injury.
