摘要:
目的  构建稳定过表达Wnt5a的K562细胞株,并观察稳定过表达Wnt5a对K562细胞生物学性状的影响。  方法  构建质粒pSEB-Wnt5a;包装得到含有逆转录病毒的上清,感染K562细胞,筛选出感染成功的细胞;Western blot检测蛋白的表达;细胞计数、瑞氏染色、透射电子显微镜观察对细胞生物学性状的影响。  结果  成功构建了质粒pSEB-Wnt5a;筛选得到了感染成功的细胞;感染pSEB-Wnt5a逆转录病毒的K562细胞Wnt5a 蛋白表达水平明显高于感染空病毒载体的细胞;与K562-vector细胞相比,K562-Wnt5a细胞的增殖被明显抑制,并出现了向成熟分化的形态学特征。  结论  成功构建了稳定过表达Wnt5a的K562细胞株;稳定过表达Wnt5a抑制K562细胞增殖并可以促进其向成熟分化,提示Wnt5a在白血病发生中可能起到抑癌基因的作用。
Abstract:
Objective   To construct the K562 cells that stably overexpresses Wnt5a protein, and to observe the effects of stable Wnt5a overexpression on the biological characters of K562 cells.    Methods   The plasmid vector pSEB-Wnt5a was constructed. K562 cells were infected by the retrovirus supernatant obtained through packaging, and the infected K562 cells were screened out. The expression of Wnt5a protein was examined with Western blot analysis. Cell counting, Wright’s staining and transmission electron microscopy (TEM) were used to observe the effects of Wnt5a overexpression on the biological characters of K562 cells.    Results   The plasmid pSEB-Wnt5a was constructed successfully, and the K562 cells infected by pSEB-Wnt5a retrovirus were screened out. Expression of Wnt5a protein in the K562 cells infected by pSEB-Wnt5a retrovirus was significantly higher than that in K562-vector cells. Compared with K562-vector cells, the proliferation of K562-Wnt5a cells was inhibited obviously, and the differentiation phenotype of K562-Wnt5a cells appeared.    Conclusion   The K562 cells that stably overexpresses Wnt5a protein are constructed successfully. Stable Wnt5a overexpression inhibits the proliferation of K562 cells and induces the differentiation of K562 cells, and it implies that Wnt5a is an antitumor factor in the pathogenesis of leukemia.
