摘要:
目的    探讨miR-1、miR-133和miR-206等3种肌肉特异性microRNAs在失神经支配所致骨骼肌萎缩中的表达变化情况。    方法    采用坐骨神经离断术建立小鼠腓肠肌失神经支配模型,采用失神经前后腓肠肌湿质量、肌纤维横截面积百分比对模型进行评价,应用Northern blot方法检测失神经支配不同时间点肌肉特异性microRNAs的表达变化。    结果    成功建立小鼠腓肠肌失神经萎缩模型,在该过程中,随着失神经支配时间的延长,骨骼肌特异性miR-206表达明显上调,miR-1、miR-133表达先快速下调后逐渐回升。    结论    随着失神经支配时间的延长,骨骼肌特异性microRNAs表达发生明显变化,骨骼肌特异性microRNAs可能在失神经介导的骨骼肌萎缩中发挥了一定作用。
Abstract:
Objective    To explore the expression changes of muscle special microRNAs (miR-1, miR-133 and miR-206) in the atrophying skeletal muscle induced by the denervation in order to establish a basis for further study on their roles in the denervated atrophying muscle.     Methods    The mouse model of sciatic nerve resection was prepared and evaluated by comparing the wet weight percentage of muscle before and after denervation and the cross section area (CSA)of the muscle fibers. Northern blotting was used to examine the levels of muscle special microRNAs expressions in the atrophying mouse gastrocnemius muscle at different time points after denervation.     Results    The mouse model of sciatic nerve resection was successfully established. The expressions of miR-206 was obviously up-regulated with the time of denervation, however, those of miR-1 and miR-133 was decreased firstly and then gradually recovered with the time elapse.     Conclusion    The changes of the expressions of the skeletal muscle special microRNAs is very striking, so we presume that they may play certain roles in the atrophying skeletal muscle resulted from denervation.
