目的 研究正常SD大鼠膀胱不同部位ICCs细胞(interstitial cells of Cajal, ICCs)的分布及其对局部逼尿肌收缩的影响。 方法 分别于正常SD大鼠膀胱顶部、体部、底部及三角区取3 mm×4 mm逼尿肌组织进行免疫荧光组织化学法检测c-kit阳性细胞的表达情况;再于上述部位取2 mm×7 mm肌条进行张力测定实验,比较各部位肌条频率及幅度的变化,分析其与ICCs密度变化的相关性。 结果 正常膀胱中存在ICCs,三角区及顶部的ICCs密度较体部和基底部明显增加,差异显著(P<0.05);三角区和顶部以及体部和基底部的ICCs密度均没有显著区别(P>0.05);各部位肌条收缩频率和幅度不同,但顶部和三角区的肌条收缩频率和幅度较体部和基底部明显增大,差异显著(P.<0.05),顶部和三角区以及体部和基底部的肌条之间则没有显著区别(P>0.05)。相关性分析结果表明逼尿肌肌条收缩频率和幅度与局部ICCs密度呈显著正相关性(P<0.05)。 结论 大鼠膀胱中存在ICCs,膀胱各部位ICCs的分布有显著差异,其与膀胱局部肌条收缩有密切关系。
Abstract:
Objective To explore the distribution of interstitial cells of Cajal (ICCs) in normal rat bladder and its effects on detrusor contraction.  Methods Detrusor tissue pieces of  about 3 mm×4 mm were obtained from the dome, triangle, lateral or base of rat bladder. The c-kit positive cells were detected by immunofluorescent staining. Detrusor strips of 2 mm×7 mm were dissected from the same sites respectively for muscle tone determination. The frequency and amplitude of the detrusor strips were compared among different sites of bladder.  Results ICCs did exist in rat bladder, and had different distribution in different sites of bladder. There were more ICCs in the dome and triangle than in the lateral or base of bladder (P<0.05), and there was no difference between the dome and triangle or between the lateral and base. The contraction of strips in different sites varied according to the distribution of ICCs. The amplitude and frequency of strips from the dome and triangle of bladder were significantly higher than those from the lateral and base (P<0.05).  The contraction amplitude and frequency of strips were positively correlated with the number of ICCs (P<0.05).  Conclusion ICCs is unevenly distributed in rat bladder, and the contraction amplitude and frequency of detrusor strips are correlated with the number of ICCs in different sites of bladder.
