目的 探讨缺氧性脑损伤后癫痫发作敏感性及其与PSD-95的关系。 方法 采用成年雄性SD大鼠,以8%氧氮混合气体建立大鼠缺氧性脑损伤模型,腹腔注射戊四唑(pentylenetetrazol,PTZ) 10 mg/(kg·5 min)和35 mg/(kg·2 d)点燃大鼠,检测其癫痫敏感性的改变。分别用免疫组织化学和免疫印迹方法观察大鼠颞叶皮层、海马和中脑神经元脱失的病理学改变以及脑内PSD-95表达的变化。 结果 缺氧损伤后大鼠癫痫敏感性明显增强。缺氧大鼠和缺氧后PTZ致痫大鼠海马CA1区、颞叶皮层和中脑神经元不同程度的脱失,缺氧大鼠脑内PSD-95表达明显降低。 结论 成年大鼠缺氧性脑损害后癫痫敏感性增强,可能与神经元脱失以及脑内PSD-95表达变化有关。
Abstract:
Objective    To explore the relationship between seizure susceptibility and PSD-95 expression in rats following hypoxic brain injury.      Methods    Male SD rats were divided into the control group and the hypoxia group. The hypoxic brain injury model was established by exposing the rats to global hypoxia induced by 8% nitrogen oxygen atmosphere. Pentylenetetrazol (PTZ) [10 mg/(kg·5 min) and 35 mg/(kg·2d)] were injected intraperitoneally into the rats from both groups to test the number of kindling, seizure stage and seizure threshold. The neuronal loss and PSD-95 expression in the temporal cortex, hippocampal CA1 subfield and midbrain were studied by using immunohistochemistry and Western blotting, respectively.      Results    The rats exposed to hypoxia exhibited increased seizure susceptibility to PTZ. Associated with this change in seizure susceptibility, neuronal loss was observed in the temporal cortex, hippocampal CA1 subfield and midbrain region in hypoxic rats and post-hypoxic kindling rats while PSD-95 expression was reduced in the both temporal cortex and hippocampus in hypoxic rats.      Conclusion    Neuronal loss is associated with the increased seizure susceptibility in adult rats following hypoxic cerebral injury. The decreased expression of PSD-95, as the consequence of hypoxic damage, may at least in part, contribute to hypoxia-induced reduction in seizure threshold.
