目的  探讨转录因子激动蛋白-2α(activator protein 2α, AP-2α)对脑外伤后蓝斑去甲肾上腺素能神经元中酪氨酸羟化酶(tyrosine hydroxylase, TH)表达的影响。 方法  将90只Wistar大鼠分为对照组和脑外伤组,脑外伤组又分为外伤后1、3、6、24、72 h共5个组(n=15)。用Feeney氏法制作机械性脑外伤大鼠模型,分别于脑外伤后各相应时间点取出脑干,对蓝斑(locus ceruleus, LC)所在部位作冷冻切片,用免疫荧光双标检测TH和AP-2的表达,观察二者在外伤后各相应时间点LC神经元中的表达变化。 结果  在脑外伤后各时段,LC神经元的TH和AP-2α 阳性细胞数量和荧光强度值均比对照组明显增加(P<0.05),3 h达到高峰,随后逐渐降低,但72 h仍然高于对照组;而且TH和AP-2α的表达变化趋势基本一致,在同一神经元可以共表达。 结论  机械性脑外伤使LC去甲肾上腺素能神经元中TH表达增加,使去甲肾上腺素(norepinephrine, NE)合成增多,NE在外伤应激过程中起着重要作用;AP-2α参与了TH的表达调控,AP-2α上调可能是导致了TH表达增加的重要原因。
Abstract:
Objective    To explore the role of transcription factor activator protein 2α (AP-2α) on the expression of tyrosine hydroxylase (TH) in noradrenergic neurons of locus ceruleus (LC) after traumatic brain injury.     Methods    Ninety rats were randomly divided into control group and injury group that comprised 5 subgroups. There were 15 rats in each group. The rat model of traumatic brain injury was established with Feeney’s methods. Brain stems were dissected from decapitated heads 1, 3, 6, 24 and 72 h after traumatic injury and freeze-mounted for cryo-sectioning. TH and AP-2α expressions in noradrenergic neurons of LC were analyzed with double immunofluorescence.     Results    The number and fluorescence intensity of TH and AP-2α-positive neurons in LC at various intervals after injury significantly increased as compared with control group (P<0.05). The expression of TH reached the peak 3 h after injury and decreased gradually, but still higher than control group 72 h after injury. The pattern of AP-2α expression was consistent with that of TH in LC, and AP-2α and TH could coexist in the same neurons.     Conclusion    The increase of TH expression in LC can synthesize more norepinephrine rapidly after brain injury. Norepinephrine plays an important role during the process of injury stress. AP-2α participates in the regulation of TH expression in noradrenergic neurons and its upregulation may be an important reason for increased TH expression in LC.
