摘要:
目的    探讨TIMP-3在移植物慢性排斥反应血管病变中的作用。    方法    在大鼠移植段主动脉转染TIMP-3质粒,利用免疫组织化学法测定移植段腹主动脉内MMP-1,2、TIMP-3表达;Northern杂交分析MMP-2表达;酶图分析术后MMP-2的活性。    结果    ①同系移植组MMP-1,2、TIMP-3表达均较低;异系移植组MMP-1,2、TIMP-3表达显著增加。②转染TIMP-3的异系移植组术后1、2周时TIMP-3免疫组化染色密度指数为(1.7±1.1)和(0.9±0.4),较单纯异系移植组、同系移植组明显增强,有显著差异(P<0.05)。③转染TIMP-3的异系移植组MMP-2活性相对单纯异系移植组、同系移植组明显降低。    结论    调控MMPs与TIMPs之间的平衡,利用TIMP-3可能有效抑制MMPs表达及活性,干预细胞外基质的形成,从而减缓CR的发生。
Abstract:
Objective    To investigate the function of TIMP-3 in the development of graft arteriosclerosis after orthotopic transplantation.     Methods    SD-Wistar abdominal aorta transplantation was performed in 60 pairs, 30 pairs of which received the exogenous TIMP-3 transfection into transplanted vessels. Wistar-Wistar abdominal aorta transplantation was performed in 30 pairs. The expressions of MMP-1, 2 and TIMP-3 in the tissues of transplanted abdominal aorta were measured by immunohistochemical method, the expression of MMP-2 was detected by Northern blot quantitative analysis and the activity of MMP-2 was observed by photo analysis on 3rd day, 1st, 2nd, 4th, 8th week after transplantation.     Results    The expressions of MMPs, TIMP-3 in the allograft group were higher than the isograft group. The expressions of TIMP-3 on 1st and 2nd week [(1.7±1.1) and (0.9±0.4)] in the allografts with exogenous TIMP-3 gene transfection were much higher than those of allografts and isografts (P<0.05), but the activity of MMP-2 was opposite.     Conclusion    While the exogenous TIMP-3 gene was admitted on early stage of CR, intervening the balance between MMPs and TIMPs, decreasing the expression and activity of MMPs, and reducing the formation of ECM can slacken CR efficiently.
