摘要:
目的    研究实验性变态反应性脑脊髓炎(experimental allergic encephalomyelitis,EAE)大鼠大脑Olig1,Olig2和Nkx2.2基因的表达规律。    方法    采用注射豚鼠脊髓匀浆的方法制作Wistar大鼠EAE模型。实时RT-PCR检测基因表达的相对值。     结果    正常大鼠大脑Olig1,Olig2和Nkx2.2都低水平表达。EAE大鼠症状发作后第6天,其表达水平达最高峰,与正常组比较,相差显著(P<0.05)。三者的变化规律一致。    结论    Olig基因表达量的增高程度可能是EAE大鼠髓鞘修复不完全的因素之一。
Abstract:
Objective    To study the experessions of Olig1, Olig2 and Nkx2.2 in th brain tissues of rats of experimental allergic encephalomyelitis (EAE).     Methods    The Wistar rats were immunized with guinea pig’s homogenated spinal cord mixed with CFA. The expressions of the three genes were detected by real-time PCR.     Results    In normal rats, Olig1, Olig2 and Nkx2.2 expressed in low level. On day 6 after onset of the symptoms of EAE, the experessions of Olig1, Olig2 and Nkx2.2 reached the peak value, with significant difference with those of normal rats (P<0.05).     Conclusion    The increasing extent of Olig1, Olig2 and Nkx2.2 is related to incomplete remyelination in EAE.
