摘要:
目的    研究FAM样酪氨酸激酶3(FLT3)靶向RNA干扰对急性髓细胞白血病细胞株THP-1的细胞增殖和周期的作用,探讨其可能的机制。    方法    体外转录合成FLT3靶向短发夹状干扰RNA(FLT3-shRNA),转染THP-1细胞;CCK-8法检测增殖抑制率、绘制增殖曲线,流式细胞法测细胞周期,半定量RT-PCR及Western blot测干扰前后cyclin D1和cyclin A的mRNA、蛋白水平的表达。    结果    FLT3-shRNA干扰可抑制THP-1细胞增殖,抑制率为(36.70±3.67)%;干扰后THP-1细胞增殖曲线滞后于对照,曲线低平缺乏典型的指数增殖特征;干扰48 h细胞G0/G1 期比例(65.51±4.48)%明显增高,S期(25.11±2.70)%下降;干扰后cyclin D1的表达下降,48、72 h对其mRNA的抑制率分别是(37.09±3.76)%、(34.30±7.60)%,对蛋白的抑制率是(37.51±1.58)%、(39.40±5.44)%,cyclin A的mRNA、蛋白的表达无变化。    结论    FLT3靶向RNA干扰可抑制THP-1细胞增殖,其机制可能与下调cyclin D1的表达有关。
Abstract:
Objective    To study the effect of FLT3-targeted RNA interfering on myeloproliferation, cell cycle of THP-1 cells and investigate the related mechanisms.     Methods    FLT3-targeted small hairpin interfering RNA (FLT3-shRNA) was synthesized by in vitro transcription system and then transfected into THP-1 cells; the growth inhibiting rates and cell growth curves were assayed by CCK-8 method, cell cycles were determined by flow cytometry, the mRNA expressions of cyclin D1 and cyclin A were detected by semi-quantitative RT-PCR and the protein expressions were detected by Western blot.     Results    The proliferation of THP-1 cells was strongly inhibited by FLT3-shRNA, with the inhibiting rate of (36.70±3.67)%. The growth curve was lagged and lack of exponential growth phase as compared to controls. Forty-eight hours after RNA interfering, the cell percentage of the G0/G1 phase [(65.51±4.48)%] was increased and the S phase [(25.11±2.70)%] was decreased. The expressions of cyclin D1 was down-regulated, of which the mRNA inhibiting rates in 48 h, 72 h were (37.09±3.76)%, (34.30±7.60)% and the protein inhibiting rates were (37.51±1.58)%, (39.40±5.44)%, but no difference of cyclin A expression was found after FLT3-shRNA treatment.      Conclusion    FLT3-targeted RNA interfering can inhibit myeloproliferation in THP-1 cells, which may be caused by the down-regulation of cyclin D1.
