摘要:
目的    研究Mash1在室管膜前下区(anterior subventricular zone, SVZa)神经干细胞向γ-氨基丁酸(gamma-aminobutyric acid, GABA)能神经元分化中的作用。    方法    体外分离培养新生0 d昆明小鼠的SVZa神经干细胞,构建Mash1与绿色荧光蛋白(green fluorescent protein, GFP)正义、反义融合蛋白表达质粒,转染SVZa神经干细胞,GFP 活体荧光标记SVZa神经干细胞,采用流式细胞仪检测在Mash1作用下SVZa神经干细胞分化为神经元的比例和分化为GABA能神经元的比例。    结果    pEGFP-Mash1-质粒、pEGFP-Mash1+质粒双酶切鉴定构建成功,核转染结果表明:GFP融合蛋白表达阳性;pEGFP-Mash1+组SVZa神经干细胞分化为神经元的比例明显高于空白对照组(P<0.05);pEGFP-Mash1-组SVZa神经干细胞分化为神经元的比例明显低于空白对照组;pEGFP-Mash1+组SVZa神经干细胞分化为GABA能神经元的比例明显高于空白对照组(P<0.05);pEGFP-Mash1-组SVZa神经干细胞分化为GABA能神经元的比例明显低于空白对照组(P<0.05)。    结论    pEGFP-Mash1-质粒、pEGFP-Mash1+质粒构建成功;Mash1可以促进SVZa神经干细胞向神经元方向分化,而且主要表现为促进SVZa神经干细胞向GABA能神经元分化。
Abstract:
Objective    To investigate the role of Mash1  in  differentiation of anterior subventricular zone (SVZa) neural stem cells to gamma-aminobutyric acid (GABA) neurons.     Methods    The neural stem cells derived from SVZa of postnatal Kunming mouse aged 0 day (P0) were separated and cultured in vitro. The rate of GABA neurons in the differentiated cells of neural stem cells of SVZa induced by different plasmids of Mash1 was detected by cell counting and flow cytometry (FCM).     Results    Double digestions verified the successful construction of Mash1-GFP+and Mash1-GFP-. The rate of GABA neuron in the group of sense Mash1 was higher than that of blank control group while the rate of GABA neuron in the group of antisense Mash1 was lower than that of blank control group.     Conclusion    pEGFP-Mash1- and pEGFP-Mash1+ plasmids have been successfully constructed. Mash1 can promote the GABA neuronal differentiation of the neural stem cells of SVZa.
